Is it possible to to block ALL web sites except for one?

  For "one" what?  For one user?

  And no, you cannot block all websites with OpenDNS.  This is not its purpose.  You'll have to unplug that cable or WLAN to the router for devices which should not have internet access.  Your router.may support such a feature too.  Check it.
